KPV (10mg) — Research Peptide
KPV (Lys-Pro-Val) is a C-terminal tripeptide fragment of alpha-melanocyte stimulating hormone (α-MSH), a 13-amino acid neuropeptide with established anti-inflammatory properties. Unlike the full α-MSH peptide, KPV does not bind melanocortin receptors with significant affinity, instead exerting its documented anti-inflammatory effects through direct intracellular mechanisms — primarily inhibition of NF-κB activation, a central pathway in inflammatory signalling.
Mechanism of Action
KPV’s primary research-characterised mechanism involves inhibition of the NF-κB (nuclear factor kappa-light-chain-enhancer of activated B cells) signalling pathway. NF-κB is a master transcription factor driving expression of pro-inflammatory cytokines including IL-1β, IL-6, IL-8, and TNF-α. KPV penetrates epithelial cells and inhibits IκB kinase (IKK) phosphorylation, preventing IκB degradation and subsequent NF-κB nuclear translocation.
Research by Dalmasso et al. (2008) demonstrated KPV’s ability to reduce inflammatory cytokine expression in intestinal epithelial cells, with effects comparable to the parent α-MSH molecule despite KPV’s lack of melanocortin receptor affinity. This suggests a receptor-independent intracellular mechanism distinct from typical neuropeptide signalling.
