Unlike the full Tβ4 protein, the TB-500 fragment retains the primary actin-binding domain responsible for Tβ4’s documented biological activities, making it the standard research form used in published studies investigating Thymosin Beta-4 biology.
Biology of Thymosin Beta-4
Thymosin Beta-4 was first isolated from thymic tissue in the 1960s and initially characterised as a thymic hormone. Subsequent research established it as a ubiquitous intracellular protein whose primary function is G-actin sequestration — binding to monomeric actin and preventing its polymerisation into F-actin filaments. This regulatory role in actin dynamics has far-reaching consequences for cell motility, as directed cell migration depends on controlled actin polymerisation at the leading edge of migrating cells.
Research Applications
Published research has investigated TB-500/Tβ4 across several biological contexts:
- Wound healing: Tβ4 promotes dermal fibroblast migration and wound closure via ILK (integrin-linked kinase) activation. Goldstein et al. demonstrated accelerated wound repair in rodent models with Tβ4 administration.
- Cardiac research: A landmark 2004 Nature study (Bock-Marquette et al.) demonstrated Tβ4 activates epicardial progenitor cells and promotes cardiomyocyte survival following myocardial injury models — a finding that generated significant follow-on cardiac regeneration research.
- Skeletal muscle: Tβ4 promotes satellite cell differentiation and muscle fibre regeneration following injury in rodent models.
